Then add 5 drops of Kovac’s reagent to the culture Note: If the Indole is negative it would be a yellow color If the Indole is positive it will equal a Red and Pink/orange color TSI Test: Triple Sugar Iron agar is used to evaluate the ability of bacteria to ferment sugars and/or to produce hydrogen sulfide. 1. Using a sterile loop, inoculate slant from the liquid culture 2. Then it is necessary to press the loop into the agar slant 3. When removing loop, smear slanted side of agar with the loop 4. Grow overnight at 37 degrees Celsius
