Microbiology
Objective:
• Cultivation of samples o Growth conditions
and equipment used
• Identification of samples o Biomedical assay
and tests available
• Evaluation of samples o Microscopy
▪ To visualize and to identify key characteristics of specimens
Procedure:
• BASIC EQUIPMENT o Cleaning
▪ Autoclave at 125 degrees Celsius
• It uses heat, pressure, and steam to sterilize
your environment
• It is not just dry air, steam is more efficient to
achieve a sterile setting
o Growing
▪ Fixed incubator: 37 degrees Celsius
• Air tight and sealed, a metal door is closed to keep the samples in a
dark surrounding
• Shelves: petri dishes that contains samples
• Tubes: contains liquid cultures
▪ Shaker incubator: 37 degrees Celsius
• When you want to take a growth media to inoculate the bacteria. Put
the growth media in the shaker and it will shake and rotate the
culture in a swirling motion to achieve growth under prime

conditions through aerating.
• Metal rings and flask is in the back
•
o Microscopy
▪ Visualizing the specimens
o Storing
▪ The refrigerator is kept at 4 degrees Celsius
• General Lab Safety o Never eat or drink in the lab
▪ This will avoid contamination risks o
Use Personal Protective Equipment (PPE) ▪
Latex or thermal gloves
• Extreme cold degree: use cold gloves
• Extreme hot degree: use hot gloves (pulling things out of the
autoclave)
▪ Eyewear protection
▪ Lab coat o Never leave the lab
while wearing PPE ▪ This includes
• Public hallway
• Bathroom
• Cafeteria
