- Amino acids substitutions are likely to be detrimental (harmful) if:
o Non-polar <=> polar
o + charged <=> - charged or vice versa
o Change involving a structurally unique residue
o Disruption of disulfide bonds
o Replacement with bulky side chain (large side chain)
