Pre-lab Questions
1. In this lab, you will conduct three different experiments that each cover the topic of selective
media and agar. How are these experiments different from one another? Explain how the
different media in each experiment are utilized in the context of selective vs differential media.
Each experimental method is designed with the objective of isolating specific bacterial types
and is guided by distinct goals. The choice of medium is influenced by its capacity to either
promote the growth of certain bacteria (selective growth) or to distinguish between bacterial
characteristics (differential differentiation). The characteristics of the microorganism under
investigation are a critical factor in selecting the appropriate medium. The principal aim is to
identify and isolate microorganisms with desired attributes. For instance, in bioprospecting
experiments, starch agar is employed to cultivate bacteria capable of starch digestion. Another
experiment focuses on isolating Gram-positive bacteria and differentiating them based on
hemolysis and mannitol fermentation using Columbia CNA Agar and Mannitol Salt Agar. In a
separate experiment, Gram-negative bacteria are selectively isolated and classified based on
lactose fermentation and colony morphology, utilizing MacConkey Agar and Eosin Methylene
Blue Agar. Each experiment utilizes media tailored to the specific requirements of the target
microorganisms, thereby enhancing the understanding of bacterial diversity and characteristics.

2. What is the difference between chemically defined and chemically complex media? Give an
example (a clinical or environmental research application) of when you would use each type of
medium to culture different microorganisms.
Chemical composition distinguishes chemically defined media from chemically complex media.
Chemically defined media, like glucose broth, are distinguished by their exact and thoroughly
recorded composition. Chemically complex media, on the other hand, such as nutritional broth,
are made up of components whose precise chemical composition is unknown and may change
from batch to batch.

3. Why do we grow microorganisms on general growth media first, before inoculating differential
media?
Transferring isolated colonies from general growth media to differential media is standard
practice because it encourages the growth of particular bacterial species. Differentiating
between the metabolic activities of different species or groups is made possible in large part by
this technique. It thus makes it possible to isolate individual colonies, guaranteeing their purity
and streamlining the subsequent examination of their overall properties.

4. What changes might you see on a differential plate that helps identify the bacteria? The
variations in color observed in the media of a differential plate or the differences in the
morphological characteristics of bacterial colonies.
