PRINCIPLES BEHIND THIN LAYER AND PAPER
CHROMATOGRAPHY:
For my practical, I tested both chromatographic methods. By
calculating their Rf values, the paper chromatography was
used to determine plant extract components, and thin layer
chromatography were utilised to identify Rf values for
various amino acids. The planar chromatography technology
known as "paper chromatography" is carried out on a specific
type of paper. The stationary phase is the paper on which the
component separation takes place.
Due to the components' different affinities for water and
solvents in the mobile phase as it moves through the pores of
the paper, the components are separated. Adsorption
chromatography between liquid and solid phases, where the
liquid phase is the mobile phase, and the solid paper is the
stationary phase. For separating various mixes comprising
polar and non-polar components, this method is quite
helpful. It is frequently used to identify various medications,
hormones, amino acids, and other substances in various
industrial applications and the pharmaceutical industry. The
thin layer of silica on its surface serves as the stationary
phase in thin layer chromatography, which is almost identical
to paper chromatography in appearance. For comparing
unknown chemicals' Rf values to published ones, these
techniques are particularly useful for identifying them. The

, attraction between each component's stationary and mobile
phases determines how each component separates. Each
component has unique qualities as a result among which
their Rf values vary. Many uses for thin layer chromatography
include the analysis of dyes in paints, inks, and fibres.
Detecting pesticides and insecticides in food products is
another use for it in the food industry. Both methods need a
solvent that can assist in moving the various components of
the sample through into the stationary phase. The solvent
then carries various components through the pores of the
stationary phase via capillary action. Each component moves
at a distinct speed, and they all absorb substances differently.
The paper is taken out and let too dry after the solvent has
reached its peak. By calculating the retention factor values
which are obtained by dividing the distances travelled by the
compound and the solvent the various components can be
quickly distinguished after drying out.
Compared to other chromatography methods, paper
chromatography is easier to use and can be done by adding
little dots of an unknown sample one at a go. After that, it is
immersed in the solvent, which raises the filter paper's
components to the top. Smaller molecules will travel more
quickly than larger ones. Due to the presence of an extremely
polar silica layer on its surface, thin layer chromatography
behaves slightly differently. The silica layer causes some
components to get more strongly adsorbed in stationary
phase, which prevents them from travelling extremely far,
while the components that are most attracted to the silica
layer migrate through the plate far. Overall, both methods
