Sheep have been dying of an unknown disease after recently being moved from winter
to summer pastures. The veterinarian has asked you to conduct a post-mortem and
submit samples to the laboratory.

1. Precautions to take before conducting the post-mortem

Before beginning the post-mortem examination, appropriate safety measures should be
followed to protect both the examiner and the quality of the samples. Personal
protective equipment (PPE), including gloves, overalls, boots, and a face mask, should
be worn to reduce the risk of infection. Clean and sterilised instruments should be
prepared to prevent contamination of specimens. The carcass should be examined as
soon as possible after death to minimise decomposition. The post-mortem should also
be carried out in a clean, isolated area to avoid spreading disease to healthy animals or
contaminating the environment. Finally, information such as the animal's history,
clinical signs, recent movement to new pastures, and suspected disease should be
recorded before the examination begins.

2. Types of samples to collect

A variety of suitable samples should be collected to help the laboratory identify the
cause of death. Fresh tissue samples from organs such as the liver, lungs, spleen,
kidneys, lymph nodes, and intestines should be collected. Blood samples, if available,
may also be useful for laboratory testing. Swabs from affected organs or visible lesions
should be taken for bacteriological examination. If poisoning or environmental
contamination is suspected, feed, water, or pasture samples should also be submitted.
Collecting different sample types improves the likelihood of obtaining an accurate
diagnosis.

3. Packaging of the samples

Samples should be packaged carefully to ensure they arrive at the laboratory in good
condition. Each specimen should be placed in a leak-proof primary container and
sealed properly. Absorbent material should be included to absorb any leakage, after
which the primary container should be placed inside a second waterproof container.
Finally, all samples should be packed in a strong, rigid outer container to protect them
during transportation. Fresh specimens must be kept cold using ice packs while
maintaining the cold chain, and all containers should be correctly labelled with the
necessary identification details. Formalin-fixed tissues should be packaged separately
from fresh samples.
