, 1. INLEIDING

1.1 DEFINIËRING MICROBIOLOGIE

= deel van biologie die micro-organismen bestudeert

• Micro-organismen: niet waarneembaar met het blote oog, microscopische afmeting
• -> soms zichtbaar onder de vorm van een kolonie of smeer (vaste voedingsbodem) of troebele
bouillon (vloeibare voedingsbodem)

In een laboratorium = aangekochte stammen met micro-organismen = American Type Culture
Collection

-> soms ook een ‘wild type’ uit een waterstaal, bloedstaal…

Verschillende domeinen microbiologie

1. Klinische microbiologie: een pathogeen micro-organisme identificeren in een patiëntenstaal
en gevoeligheid ten opzichte van antibiotica bepalen
2. Milieumicrobiologie: kwaliteit van water controleren op aanwezigheid van micro-organismen
3. Voedingsmicrobiologie: voedingswaarden testen op aanwezigheid van micro-organismen die
verantwoordelijk kunnen zijn voor infecties of voedselvergiftigingen
4. Onderzoekslabo’s: met micro-organismen werken om ze genetisch te manipuleren of typeren

1.3 PLAATS VAN HET MICRO-ORGANISME IN DE NATUUR


1.3.1 PROTISTA
= ééncelligen of meercellige organismen zonder gedifferentieerde weefsels

• Basis van micro-organisme = cel met DNA, RNA, proteïnen, lipiden


1.3.2 HET 3RIJKENSYSTEEM
Ernst Haeckel: indeling op basis van microscopische en macroscopische kenmerken

1. Planten: fotosynthese, grote uitwendige oppervlakte, celwand, geen actieve beweging
2. Dieren: intestinale tractus, grote inwendige oppervlakte, geen celwand, actieve beweging
3. Protista: bacteriën, schimmels/gisten, wieren, ééncelligen dieren (protozoa)

1. Prokaryoten: oudste cel, primitief, geen celkern (DNA los in cel), geen celorganellen (wel
ribosomen), voortplanting door amitose (dwarse deling)
2. Eukaryoten: celkern met chromosomen, bezit celorganellen, voorplanting door mitose of
meiose
