Mutton in flasks was boiled and then covered. Broth became cloudy and contained microorganisms. Argued for spontaneous generation. (Didn't boil flask long enough.) Lazzaro Spallanzani (SG) (1765) Again boiled broth in two flasks and then capped one. Growth in uncapped flask, not in capped. Louis Pasteur (SG theory) (1861) Confirmed Tyndall's findings. Placed nutrient solution in flasks. Created flasks with long, curved necks. Boiled solutions. Left flasks exposed. No micro growth. Disproved spontaneous generation. John Tyndall (SG) (1877) Final blow to SG theory. Demonstrated that dust carries microorganisms. If dust is absent, nutrient broth remains sterile, even if directly exposed to air. Also provided evidence for existence of exceptionally heat resistant forms of bacteria. Joseph Lister (disease) Antiseptic surgery. Provided indirect evidence that microorganisms were the causal agents of disease; developed a system of surgery designed to prevent microorganisms from entering wounds; had patients with fewer post-op infections. Louis Pasteur (disease) Germ Theory of Disease. Showed that pébrine disease of silkworms was caused by a protozoan. Showed that fungi cause diseases of silkworms. Robert Koch (disease) Established relationship between Bacillus antracis and anthrax. Used criteria developed by teacher Jacob Henle. These criteria are now known as Koch's postulates, which are still used today to establish link between diseases and microorganisms. Koch's Postulates 1. The microorganism must be present in every case of the disease, but absent from healthy animals. 2. The suspected microorganism must be isolated and grown in a pure culture. 3. The same disease must result when the isolated micro is inoculated into a healthy host. 4. The same micro must be isolated again from the diseased host. Where were microbial cultivation techniques developed? Koch's lab. Petri dish. Agar suggested by Walter Hesse's wife, Fannie Eilshemius. Pasteur (vaccination) Chicken cholera experiment. Edward Jenner (vaccination) 1798. Used a vaccination procedure to protect individuals from smallpox. (Preceded the work of establishing the role of micro in disease.) Pasteur and Roux Discovered that incubation of cultures for long intervals between transfers, caused pathogens to lose their ability to cause disease. Pasteur and coworkers developed vaccines for chicken cholera, anthrax, and rabies. Paul Ehrlich (chemo) Chemotherapy. Developed arsenic-based drug, Salvarsan, that was effective against syphilis. It was the first drug methodically developed to treat a specific disease. Gerhard Domagk (chemo) Prontosil discovered. Protects from lethal staphylococci and haemolytic streptococci. Discovery of the antibacterial action of the sulfonamides. Alexander Flemming (chemo) Discovery of penicillin. D. Iwanowski (viruses) Discovered tobacco mosaic viruses. Took samples of plant, blended, and filtered. (Virus=poison) Beadle and Tatum (modern) (1940s) Microbial genetics. Worked with fungus, neurospora. Avery McLeed and McCarthi (1944)Discovered DNA is genetic material using S. pneumonia. Berg, Gilbert, and Sanger (1980)Developed recombinant DNA technology Milstein, Kohler, and Jerne (1986) Monoclonal antibodies
