Ex: lactose intolerant, because they aren't making lactase to breakdown the lactose. So drinking lactose increases the production of lactase to break it down faster. Metabolizing it too fast= tolerant to that enzyme/drug. Inhibition: high plasma levels -drug inhibits the enzyme that breaks down so it increases the drug levels and increase side effects and plasma levels. Genetics: some people have slow breakdown or fast Disease: if liver is compromised, it may not breakdown drug effectively and drug levels will rise.;High Extraction Drugs - (first order metabolism: liver first!) Flow dependent: limiting factor is blood flow.;Low Extraction Drugs - Zero order drugs
