 Ans: Blood agar is an enriched and differential media used for the growth,
differentiation, and detection of organisms with hemolytic activity. Hemolysis is the
destruction of red blood cells. Some organisms produce enzymes known as hemolysins
in order to break open red blood cells and use the contents, such as hemoglobin, as a
nutrient source. Streptococci are the most common hemolytic bacteria that cause
disease and much of their virulence can be attributed to their hemolytic activity.
Streptococci are even classified according to their hemolytic activity, such as Group A
Strep (GAS) or Group B Strep (GBS). This is known as the Lancefield classification system,
named after Rebecca Lancefield.

Hemolytic activity by bacteria occurs in three forms; alpha, beta and gamma. Alpha
hemolysis results in lysing open of red blood cells and destruction of hemoglobin, but iron
and other cell remnants are left behind. Beta hemolysis is complete destruction of the red
blood cell and no waste is left behind. Gamma hemolysis is the absence of any hemolytic
activity. By adding red blood cells to an agar base, we can easily determine if an organism
is hemolytic and which type of hemolysis is occurring. This is used to classify organisms,
for identification and detection of virulence.

The organisms used in this experiment are pathogenic, so be sure to use universal
precautions when handling them.
