“Pre-Lab Questions”
1. Which DNA nitrogenous bases pair with each other? Which bases are purines, and which are
pyrimidines?
For DNA nitrogenous bases, adenine pairs with thymine and cytosine pairs
with guanine. Adenine and guanine are both purines, and cytosine and
thymine are both pyrimidines.
2. How is DNA information used to make proteins? What are the steps of this process?
Proteins are made from DNA through translation and transcription. DNA is
transcribed into mRNA with the use of RNA polymerase. When mRNA
reaches a ribosome, tRNA translates new proteins from the mRNA.
3. Give an example of a scenario in which you would perform PCR vs a scenario in which you would
use recombinant DNA technology.
An example of when you would use PCR is if there is DNA at a crime scene
and you would need to amplify the sample by making copies of it. An
example of when you would use recombinant DNA technology is when you
would want to make a genetically modified food.
4. What occurs during each of the three steps involved in the PCR cycle? How has the use of PCR
changed biotechnology?
During denaturing, the mixture is heated to just below boiling temperature
