How do you prepare a heat-fixed smear of proper - Place a drop of DI (deionized) water on a clean slide
density for staining a solid, slant culture? - Aseptically obtain a sample of bacteria on loop and mix into drop of
water
- Spread liquid over a small area of the slide (coin sized)
- Place slide on slide warmer (if air drying - set out at room temp)
- Once smear is completely dry, pass over flame 3 times to heat fix
What are the steps for preparing a Gram Staining - Heat-fixed emulsion slide
Procedure - 1 drop of crystal violet (primary stain) for 45 seconds
- Gently rinse with distilled water into Tupperware container
- Cover smear with Iodine stain (mordant) for 15 seconds
- Gently rinse slide with distilled water
- Trickle down slide with 95% ethanol (decolorizer) until run off is clear
- Gently rinse with distilled water
- 1 drop Safranin (counterstain) for 30 seconds
- Rinse with distilled water
- Gently blot dry in a tablet of bibulous paper
What color is a gram-positive reaction? purple/blue
