Principe fluorescentie microscopie

With a fluorescence microscope, the fluorescent dyes are made visible. The light comes from a powerful source that is passed through two filters. One to filter the light before the sample is reached (excitaKon filter) and one to filter the light obtained from the sample (emission filter). The first filter allows only the wavelengths that excite the specific fluorescent dye to pass through, while the second filter blocks this light and allows only those wavelengths that are emi^ed when the dye fluoresces to pass through. Fluorescence microscopy is usually used to detect specific proteins or other molecules in cells and Kssues. A commonly used technique is to link fluorescent dyes to anKbodies, which then selecKvely bind to specific proteins in cells or in the ECM. Two fluorescent dyes commonly used for this are fluorescein, which emits a green fluorescence a[er excited with a blue light, and rhodamine, which emits a deep reed fluorescence a[er excitaKon with green- yellow light. By linking one anKbody to fluorescein and the other to rhodamine, you can compare the distribuKon of different molecules in the same cell. Note that it is not possible to observe both at the same Kme.
