Analytical Chemistry III Final Examination with Rationale. VERIFIED. First Semester SY 2023/2024:- CEBU NORMAL UNIVERSITY College of Arts and sciences Answer briefly. 1. Define the following: a. Elution- The chromatographic process of using a solvent to extract an adsorbed substance from a solid adsorbing medium. The removal of antibody from the antigen to which it is attached. b. Mobile phase- The mobile phase is part of gas chromatography mass spectrometery testing. It is an an inert gas that the sample is injected into that will carry it through the stationary phase, it moves the sample so that it is mobile. The analysis of the sample solution takes into account the interactions between these two phases as the mobile phase moves through the stationary phase. c. Stationary phase The stationary phase remains fixed in place. The stationary phase acts as a constraint on many of the components in a mixture, slowing them down to move slower than the mobile phase. d. Partition ratio The partition ratio, also known as the capacity factor (k') is the amount of time a compound remains in the stationary phase relative to the mobile phase. e. Retention time Retention time (RT) is a measure of the time taken for a solute to pass through a chromatography column. It is calculated as the time from injection to detection. The RT for a compound is not fixed as many factors can influence it even if the same GC and column are used. f. Retention factor The amount that each component of a mixture travels can be quantified using retention factors (Rf). The retention factor of a particular material is the ratio of the distance the spot moved above the origin to the distance the solvent front moved above the origin. 2. What is the difference between gas-liquid and liquid-liquid chromatography? The main difference between gas and liquid chromatography is that the mobile phase of gas chromatography is a gas, which is mos
