MOUNTING - LABELLING
Process of applying a mounting medium between the stained tissue section and the coverslip. Mounting
medium/mountant should have a refractive index close to that of the slide which is 1.518 (yung mountant
dapat kasing clear ng coverslip at ng slide)
Purpose/Why mounting is done?
a. For permanent safekeeping
b. To protect tissue from damage/injury
c. To facilitate easy handling & storage
d. To prevent bleaching of sections
2 TYPES OF MOUNTANT
1. RESINOUS MOUNTANT
2. AQUEOUS MOUNTANT
Resinous mountant
o Used for preparations that have been dehydrated and cleared with xylene.
o PERMANENT mountants/mounting media
o I.e.
CANADA BALSAM Resinous natural mountant
The mountant that is commonly used
Refractive index: 1.524
Extracted from Abus Balsamea
Disadvantages:
1. It darkens with age
2. It causes gradual fading of many stains
DPX Refractive index: 1.532
XAM Refractive index: 1.52
CLARITE Refractive index: .544
AQUEOUS MOUNTANT
o Mounting media used to mount water miscible preparations
o Water is an example of aqueous mountant
o Semi-permanent/temporary mounting media/mountants
o I.e.
GLYCERIN JELLY Refractive index: 1.47
aka Kaiser’s
FARRANT’S Refractive index: 1.43
aka Gum arabic
APATHY’S Refractive index: 1.52
Used for methylene blue stained nerve preparations
BRUN’S FLUID Recommended for mounting frozen sections directly from water
