Title: PL01, Keeping a Lab Notebook (Initials, Lab Number, Lab Title)
Objective: To establish an organized template for keeping experimental records, procedures and results.
Procedure:
1. This is where each step of protocol is recorded. . (chemicals used, temperature, growth conditions, plates etc.)
2. Clearly indicate deviations that may occur during the experiment in RED font
Notes: Additional helpful information.
Course Objectives:
• Cultivation of samples (growth conditions to study pathogens in lab settings) – Equipment used to cultivate
bacteria
• Identification of samples (biochemical assays- how to determine type of bacteria or pathogen a patient has) –
Tests available
• Evaluation of samples (microscopy) – Visualization and recognition of key characteristics of pathogens
Equipment:
1.) Cleaning: Main tool is Autoclave uses heat, pressure and steam to sterilize environment, chamber with thick
door, Used for cleaning tools to be used in lab or to kill something, at least above 125 deg C, *Not just dry air,
Steam is more efficient than hot dry air, sterile condition in a few minutes (would take several hours with hot air),
also hot steam comes out when done so it must be depressurized and opened slowly.

2.) Growing: 1- Fixed incubator 37 deg C chamber, petri dishes inside contain samples, tubes hold liquid cultures,
door is air tight and sealed and when shut keeps dark conditions
Growing 2 Shaker incubator 37 deg C), Take growth media (liquid), inoculate bacteria, shakes and rotates
bacteria in swirling fashion to help aerate it and help bacteria grow under optimal conditions

4.) Storing (Refrigerator 4 deg C to slow bacterial growth so samples can be kept for long term); may contain test
tubes with nutrient broth and liquid culture, also petri dishes wrapped in parafilm (keeps it air tight), also test tube
racks
SAFETY:
1.) Never eat or drink in the lab (contamination risk),
2.) Use PPE (personal protective equipment)
Latex Gloves (more common), Nitrile Gloves (if allergic to Latex, purple, blue or green), Thermal Gloves for extreme
Cold, Thermal Gloves for extreme Heat (pulling things our of autoclave)
Eye ware protection
Lab coat – protection from being spilled on
3.) Never leave lab while wearing PPE (public hallway, bathroom, cafeteria)
Results: Summary of final outcome of the experiment. Main point of what was done and the outcome

Title: PL02 Basics of Microscopy
Objective: To become familiar with basic components of light microscope as well as how to load sample for viewing.
Procedure: 1- Review parts and components; 2- Load sample slide onto microscope; 3- Select magnifications; 4- Make
necessary adjustments to optimize sample visualization
Notes: Microscopy is the same as using a magnifying glass; 1. Lens magnifies object, 2. Need object to look at, and 3.
Need light source.
